When keeping fish for consumption, (ie. flounder, striped bass) is it better to kill them quick or just throw them in the cooler and let them expire? What would be the best way to kill them quickly?

Unquestionably the best way to "kill them quick" and make for the best table fare is to bleed them. Surest way to bleed a fish is to cut the gills. Even flounder will taste better when bled as soon as they are caught.
